Abstract Background Prestin (SLC26A5) is responsible for acute sensitivity and frequency selectivity in the vertebrate auditory system. Limited knowledge of prestin from experiments using site-directed mutagenesis or domain-swapping techniques after amino acid residues were identified by comparing sequence to those its paralogs orthologs. Frog only representative amphibian lineage studies it quite rare with one species identified. Results Here we report a new coding SLC26A5 frog species, Rana catesbeiana (the American bullfrog). In our study, gene has been mapped, sequenced cloned successively RNA-Seq. We measured nonlinear capacitance (NLC) both hair cells ’s inner ear HEK293T transfected this gene. expressing showed electrophysiological features similar that ear. Conclusions mapped cDNA The 2292 bp long, encoding polypeptide 763 residues, 40% identity mammals. This could encode active protein conferring NLC HCs mammalian cell line. While orthologs, evolutionarily changing function becomes more advanced than avian teleost prestin.
